This talk will cover the fundamental principles of optical microscopy, microscope components and settings. It will briefly review how optical microscopes work, and the effect that microscope components and settings can have on measurement accuracy. Practical examples will be given of how microscope and camera settings create error in measurement results, and best practice recommendations given. The principles of use for digital cameras and image acquisition will also be discussed. All of these factors become important when we consider the use of automated image analysis and machine learning, as the quality of acquired images and measurement data may significantly effect the success of automation.
